Characterization study of nasal deposition targeting the Central Nervous System

Nasal drug delivery has gained increasing interest for central nervous system targeting via nose-to-brain pathways, primarily through the olfactory and trigeminal nerves. Nasal powders offer advantages over liquid formulations, including improved API stability and prolonged residence time, but their performance assessment is limited by the lack of standardized deposition methodologies. 

This study developed an in vitro analytical framework to evaluate nasal powder deposition using two distinct nasal casts—an aluminum Alberta Idealised Nasal Inlet and a 3D printed polymeric cast—highlighting the strong influence of particle size, device, and nasal geometry on regional deposition. The results underscore the need for standardized nasal models, as cast selection markedly impacts olfactory targeting and formulation screening outcomes. Biopharmaceuticals are a growing segment in Pharma with still significant formulation challenges like viscosity of high concentration forms, and stability. Traditional excipients often fall short, driving the need for next-generation solutions. Hovione will be presenting a library of novel synthetic sugars that have enabled notable improvements in viscosity reduction and stability, putting forward a rational approach towards finding the best excipient-protein combination.

This development arrives as intranasal delivery gains traction for systemic, CNS and rapid-onset therapies.
