Optimization of a Roller Compaction Process for a Spray Dryer Formulation Using Powder Rheology

Spray drying has become an established technique in the pharmaceutical industry for the production of amorphous solid dispersions from crystalline Active Pharmaceutical Ingredients (APIs) to overcome bioavailability challenges. The spray dried dispersion (SDD), containing both the API and the stabilizing polymer matrix may correspond to a significant fraction of the final formulation, leaving little flexibility for adjusting the blend properties with the addition of excipients and often requiring the use of dry granulation by roller compaction to improve powder properties. In this work we exemplify the development of a dry granulation process for and SDD supported by powder rheology.
